Title: howto configure a donotcall list for incoming and manual outgoing call?

I have a customer with Genesys SIP Server and They want to have a donotcall list for incoming and manual outgoing call. A list contains around 50 phone numbers. Plz help me how to do that

Incoming? Inbound? You can't block numbers by a simple configuration. You need to modify your strategy in order to seek those numbers somewhere and then route them to a disconnect function.
By somewhere means what ever suits your customer needs better, a DB, a simple List at IRD, etc. Because you will also manage it and add, delete numbers.
For Outbound, if using OCS you can check the do not call feature from OCS itself.
If manual, then you can do something at your Softphone or if using WDE you can use dial plan rules.
So, no simple click to go in order to solve your question

Just to add some thoughts for manual Outbound - you can also configure the dial plan at the Extension DN level so you can set it up even without WDE.

Thanks a lot cavagnaro. The simple way for our Customer is using dial-plan  :)
